The synthesis core is responsible for providing the Research Projects and the other two Research Support Cores with test compounds and analytical standards. The Specific Aims of this Research Support Core are the preparation of analytical standards for the Analytical Core (Aim 1), the preparation of an airborne PCB mixture for in vitro and in vivo studies (Aim 2) as well as the synthesis of individual PCB congeners (Aim 3), methoxylated and hydroxylated PCB metabolites (Aim 4), quinoid metabolites of PCBs and their glutathione conjugates (Aim 5), and PCB sulfates (Aim 5). Since funding began in March of 2006, this Core has synthesized, purified and characterized numerous PCB congeners and metabolites. These compounds – some in large quantities – were made available to four of the six Research Projects of the University of Iowa Superfund Research Program for in vitro and in vivo studies. Ongoing synthetic efforts by the Research Support Core focus on the synthesis of metabolites of PCB 3 for plant metabolism and genotoxicity studies (PCBs: Metabolism, Genotoxicity and Gene Expression in Vivo and Phytoremediation to Degrade Airborne PCB Congeners from Soil and Groundwater Sources), hydroxylated PCB metabolites and PCB sulfates for structure-activity studies (PCBs and Hydroxysteroid (Alcohol) SuIfotransferases), individual PCB congeners for phytoremediation studies (Phytoremediation to Degrade Airborne PCB Congeners from Soil and Groundwater Sources), diazomethane as a derivatization agent (Analytical Core in support of the Atmospheric Sources of PCB Congeners project and Airborne Exposure to Semi-volatile Organic Pollutants) and the preparation/maintenance of an airborne PCB mixture for inhalation toxicology studies (Inhalation Toxicology Core).
